How to Travel from Hong Kong to Shenzhen
Hong Kong and Shenzhen face each other across the Shenzhen River. The fastest way to travel from Hong Kong to Shenzhen is taking a high speed train, which only takes around 20min with frequent departures daily. In addition, tourists can take MTR/subway or city bus to arrive at Luohu Port, Huanggang Port and Futian Port first, then pass the border and enter Shenzhen. Flexible buses have many departures and arrival stations in the two cities, which provides more choices for tourists. Direct ferries are also a feasible way, operating from HKIA Skypier or HK Sheung Wan Ferry Terminal to Shenzhen Shekou or Fuyong Port.
Via Luohu Port
→ Take Hong Kong MTR East Rail Line to Lo Wu Station.
→ Follow the signs to reach Hong Kong Customs.
→ Get through customs and cross the border, then arrive at Shenzhen.
Via Futian Port
→ Take Hong Kong MTR East Rail Line or Bus No. B1 to Lok Ma Chau Station.
→ Follow the signs to Futian Port.
→ Clear the customs and cross the border to reach Shenzhen.
Via Huanggang Port:
Take Bus No. 605, 76K or B1 and get off at Huanggang Port. Get off there and clear the Hong Kong Customs to reach Shenzhen.
Ferries to/from SkyPier are only available for airline travellers at HK International Airport.
1. Bus: Take a direct coach from busy places in HK like Plaza Hollywood, Causeway Bay Times Square/ Prince Edward/ Tsuen Wan Nina Tower/ Ocean Park/ Disneyland Park ... to Shenzhen Bao'an Airport: 1-2h, HKD100-250
2. High Speed Train + Metro:
→ Take a high speed train from West Kowloon Railway Station to Shenzhen Futian Railway Station: 20min, CNY68 for a second class seat
→ Take metro line 11 to Bao'an Airport Station: 30min, CNY7
3. Ferry + Metro:
→ Take a ferry from HK Sheung Wan Terminal to Shekou Port: 55min, CNY90-150
→ Take metro line 2 to Houhui Station, then transfer to metro line 11 to Bao’an Airport Station: 20min, CNY6

No, you don't need a visa. Singaporeans are granted 30-day visa free entry to Mainland China.

You can apply for a China visa at HK visa service center center at 20/F, Capital Center, 151 Gloucester Road, Wan Chai, Hong Kong.

What are my travel options (time, cost, and visa requirement) if I want to do a day trip to Shenzhen from HK? I want to find the quickest but most efficient route. I previously thought the 20 min bullet train would be best, but the Shenzhen North Railroad doesn't seem to be an approved port for the 144 hour visa free option. It seems like my best option is to do the ferry from Sheung Wan to Shekou Port as Shekou is an approved port for the 144 hour visa free option.

I think you can take HK MTR to Luohu (Lo Wu), or Huanggang Port (Lok Ma Chou in HK), or take a ferry to Shekou to get a Shenzhen VOA at a cost of CNY 130. To utilize the 144, you need to leave for another country or region except for HK after visiting Shenzhen to make it a transit route.

About option 3, one thing added: cannot go back to HK; a transit route HK - Shenzhen - the third country or region is a must.
West Kowloon Railway Station is not an eligible port to arrive.
You mean take a ferry from HK to Shenzhen then return to HK by high speed train under the 144. Sorry, it's not an eligible transit route but a return trip.
